Cylinder 3 misfire

Which cylinder is number 3 on a 2000 Tacoma 3.4 liter? I have a cylinder 3 misfire?

Thanks, I can't find my Haynes manual. Probably a coil pack, I'm assuming.

If it misfires under load than it tends to be a bad coil.

The cylinders on a 3.4 V6 Tacoma go 1-3-5 on the right passenger side with 1 being closest to the front end, and 2-4-6 on the left driver side with 2 being closest to the front end. Cylinder 3 is the middle cylinder on the passenger's side, which is the side that houses the electronic coils for firing order. Unfortunately, coils are expensive, but sfstud2002 is correct in his or her (LOL) analysis of your dilemma. I am sure your problem will be rectified with a new coil. You should probably replace the spark plugs while you are at. Good Luck.
